We are studying the population diversity of human immunoglobulin regions. The 1000 genome and similar large scale human sequencing projects are endeavouring to more fully understand the differences within and between populations at the level of sequence variation, both for coding and non-coding regions. The stated aim of the 1000 genomes project is to catalogue variations with frequencies down to 1% within a population. We are seeking to understand this variation as it applies to immunoglobulin coding regions including those of the heavy chain and both light chain genes. As a base reference sequence, we’ve chosen to deep sequence cloned BACs which encompass the 3 human immunoglobulin loci. We propose to use the Illumina platform to complete this deep sequence using fairly large redundancy and pair-end sequences.
