This study employed a mouse model using sham-operated and ovariectomized mice, with or without high-fat diet-induced obesity, to disentangle the roles of menopause and obesity. Ovariectomized mice on a high-fat diet were further treated with estradiol to assess the regulatory effects of hormone supplementation on the gut microbiota. Supplemental Table 1 listed microbial ASVs signficantly affected by high-fat diet. Supplemental Table 2 listed microbial ASVs signficantly affected by ovariectomy. Supplemental Table 3 listed predicted microbial pathways affected by high-fat diet, Ovx, or Estrogen supplementation. Supplemental Table 4 listed Effects of estrogen on the gut microbiota at the ASV level in Ovx_HFD_Estrogen versus Ovx_HFD groups. Supplemental Figure 1 showed impact of obesity, ovariectomy, and estrogen supplementation on the Firmicutes: Bacteroidota ratio. Supplemental Figure 2 showed effect of estrogen supplementation on body composition in ovariectomized females.
