The SDS-sedimentation volume (SSV) is a critical indicator for assessing wheat gluten quality and is widely used in evaluating wheat processing quality. However, the molecular mechanisms regulating SSV remain poorly understood. In this study, We cloned the TaSED gene via map-based cloning, which is a positive regulator of SDS-sedimentation volume (SSV), and its function was validated through knockout and overexpression lines. To further explore the TaSED regulatory role on SSV, we performed RNA-seq analysis on grains 21 DPA from knockout line tased#1, RNAi line taspa#5, and overexpression line TaSED-OE#3. The expression of HMW-GS and gliadin genes was downregulated in the tased#1 and taspa#5 but upregulated in TaSED-OE#3. Our findings provide new insights into the molecular mechanisms governing SSV.
