This study leverages ATAC-seq to reveal Arid3a's chromatin accessibility effects in VSMCs, identifying key gene expression changes and implicating Arid3a in actin filament dynamics linked to AAA pathogenesis. We utilized RNA interference technology to knockdown Arid3a in Vascular Smooth Muscle Cells (VSMCs), successfully establishing several Arid3a knockdown cell lines. In this study, we applied ATAC-seq to dissect the chromatin accessibility dynamics in Vascular Smooth Muscle Cells (VSMCs) following Arid3a knockdown. The ATAC-seq assay was designed to capture the open chromatin regions that are potentially bound and regulated by Arid3a, offering insights into its direct and indirect transcriptional regulatory effects.
