RNA was isolated from HSC 4 cells after treatment with peptides SSTP1 and SSTP 2 using RNeasy mini kit (Qiagen) as per the manufacturer’s instruction and reverse transcribed to double-stranded cDNA. RNA-Seq analysis was done on Illumina platform to identify the target genes on treatment with SSTP1 in comparison with SSTP2, in HSC4 cells. Out of the 34 to 37 million reads obtained, 31 to 34 million reads per samples were mapped on the indexed Human reference genome GRCh38.p7. 208 up-regulated genes and 118 down-regulated genes were identified through differential gene expression analysis.The RNA-Seq results were validated by 12 up-regulated and four down-regulated genes by RT-PCR and q-PCR.
