A test of the accuracy of taxonomic assignments of some <i>CO1</i> sequences was built into the technical validation process of MIDORI2. We removed 186 complete mitochondrial <i>CO1 </i>gene sequences, one from each of the 186 taxonomic classes represented in MIDORI2, from the LONGEST_GB242 <i>CO1</i> database to use as queries.Accuracy tests were conducted for three regions of the query sequences: 1) the complete mitochondrial <i>CO1</i>, 2) the ~658 bp <i>COI </i>“barcode region” (Folmer et al., 1994), and 3) a ~313 bp fragment (Leray et al., 2013) within the barcode region.<br>The files contains <i>CO1</i> sequences of the MIDORI2 database. The 186 mitochondrial <i>CO1 </i>gene sequences, that were used as query to compare the performance of five taxonomic assignment programs (Qiime2, Mothur, RDP classifier, Sintax and BLAST+), were removed from these files.
