本发明公开了一种应拉木胶质层显微切片 的染色方法,包括以下步骤:(1)将阔叶树材倾斜或弯曲的树干或树枝锯成圆盘,确定应拉木区域;(2)在所述步骤(1)确定后的应拉木区域选取木样,经水煮排气、软化后,切取横切面切片置于蒸馏水中洗;(3)将步骤(2)洗涤后的切片立即用番红‑星蓝乙醇染液或番红‑星蓝乙酸染液双重染色进行,染色过程中用梯度乙醇溶液进行脱水,得到脱水染色后的应拉木胶质层显微切片。该染色方法使用番红‑星蓝染液对应拉木进行染色,可清晰地将应拉木胶质层与相邻壁层区分开来,便于观察胶质层的微观结构,更有利于对图像按照饱和度和灰值进行处理和分析,为研究应拉木提供更多可行性。

This invention discloses a staining method for microscopic sections of tension wood gelatinous layers, which includes the following steps: (1) Saw the inclined or bent trunks and branches of broad-leaved trees into discs, and identify the tension wood regions; (2) Collect wood samples from the tension wood regions identified in step (1), boil them for degassing and softening, then cut transverse sections and rinse them in distilled water; (3) Immediately perform double staining on the washed sections from step (2) using safranin-star blue ethanol staining solution or safranin-star blue acetic acid staining solution, and use gradient ethanol solutions for dehydration during the staining process to obtain the stained and dehydrated microscopic sections of tension wood gelatinous layers. This staining method uses safranin-star blue staining solution to stain tension wood, which can clearly distinguish the gelatinous layers of tension wood from adjacent wall layers, facilitating the observation of the microscopic structure of the gelatinous layers. It is more conducive to the processing and analysis of images based on saturation and gray values, providing more feasibility for tension wood research.
