Primary human foreskin fibroblasts (HFF) were infected with wild-type HCMV strain TB40 or BAC2 at a multiplicity of infection (MOI) of 10. Cells were treated with 4sU 1h before harvesting. Sequencing libraries were treated with IAA and further prepared using the dRNA-seq and STRIPE-seq protocol from the indicated timeoints of infection Overall design: HFF were infected with HCMV strain TB40 or BAC2. Cells were treated with 4sU as extracted RNA with IAA using the SLAM-seq protocol. Transcription start sites were enriched using the dRNA-seq and STRIPE-seq protocol on total cellular RNA. Two independent biological replicates were analysed.
