YM155 in combination with EZH2 inhibiton induces synthetic lethality

EZH2 inhibitors have been developed to treat certain cancers. To maximize the anticancer acttivity of EZH2 inhibition, we combined an EZH2 inhibitor EPZ6438 with YM155, which leads to dramatic cell death. We used microarrays to detail the global gene expression profiles. BE2C cells treated with either 5uM EPZ6438, 5nm YM155 or combination of EPZ6438 and YM155 for 8hours and 12hours. RNA were extracted by Rneasy kit from QIAGEN, which were subject to Affymetrix hybridization with GeneChip™ Human Gene 2.0 ST Array.
