Chromatin immunoprecipitation DNA-sequencing (ChIP-seq) for Pol κ in HEK293T cells. Overall design: Genome editing-based integration of three tandem repeats of the FLAG epitope tag (3×FLAG tag) into endogenous Pol κ was conducted. Then we crosslinked DNA and proteins in chromatin, fragmented DNA, immunoprecipitated endogenous Pol κ and its associated genomic DNA using anti-FLAG M2 beads, and subjected the resulting DNA fragments for next-generation sequencing analysis.
