RNA profiling of MPNST xenografts treated with anti-endoglin (ENG) therapies

In order to analyze the genes and molecular pathways modulated by ENG targeting in MPNST, we performed RNA-seq on STS26T xenografts treated with TRC105/M1043 and compared their expression profiles with those of IgG-treated control tumors. Overall design: 1x10^6 STS26T cells were subcutaneously injected into athymic nude mice and tumors were allowed to develop for one week until they were palpable (~100 mm3 average). Then, mice were treated with either the combination of anti-human and anti-mouse monoclonal antibodies (TRC105 and M1043, respectively) or the corresponding IgG control. After three weeks of treatment, mice were sacrificed and tumors were dissected. RNA was extracted with trizol and subsequent Qiagen columns. RNA sequencing was performed afterwards.
