The goals of this study are to compare Next-Generation-Sequecing (NGS)-derived genome-wide occupancy of TAF (Taf1) comparing with other basal initiation components (TBP and TFIIB), histones (H3, H4, Htz1 and H4 acetylation) and histone regulator complexes (Swr1, Bdf1) in S. cerevisiae. Overall design: Examination of H3, H4ac, TBP, Bdf1 (SRP010134), Taf1 (SRP010134), Swr1 (SRP028548/PRJNA214015), Sua7 (SRP036647/PRJNA237486) and Htz1 (SRP035413/PRJNA235191) in normal growing cells. Examination of Taf1 and TBP (Spt15) in Rpb1- and Bdf1- nuclear depleted cells.
