TP53-TAU axis regulates microtubule bundling to control alveolar stem cell mediated regeneration

To assess the morphological dynamics during the differentiation of alveolar epithelial type 2 cells (AT2s) into alveolar epithelial type 1 cells (AT1s), we established an ex vivo differentiation model in 2-dimensional cultures with defined conditions using purified AT2s from mouse lungs. Overall design: Purified murine AT2s were seeded on collagen-1 or fibronectin with serum-free feeder-free medium. We performed bulk RNA sequencing on cells collected from collagen-1 (for AT2s maintenance) or fibronectin-coated plates harvested on days 5 (for alveolar transitional cell states induction) and days 9 (for AT1s induction).
