The northern tree shrew (Tupaia belangeri) possesses high potential as an animal model of human diseases and biology, given its genetic similarity to primates. Although genetic information on the tree shrew has already been published, some of the entire coding sequences (CDSs) of tree shrew genes remained incomplete, and the reliability of these CDSs remained difficult to determine. To improve the determination of tree shrew CDSs, we performed sequencing of the whole-genome, mRNA, and total RNA and integrated the resulting data. Additionally, we established criteria for the selection of reliable CDSs and annotated these sequences by comparison to the human transcriptome, resulting in the identification of complete CDSs for 12,612 tree shrew genes and yielding a more accurate tree shrew genome database (TupaiaBase: http://tupaiabase.org ).
北树鼩(Tupaia belangeri)因与灵长类动物具有较高的遗传相似性,作为人类疾病与生物学研究的动物模型具有极高应用潜力。尽管树鼩的遗传信息已公开发表,但部分树鼩基因的完整编码序列(coding sequences, CDSs)仍不完整,且此类CDSs的可靠性难以判定。为提升树鼩CDSs的验证与注释质量,本研究对全基因组、mRNA及总RNA进行测序并整合所得数据;同时建立了可靠CDSs的筛选标准,通过与人类转录组比对完成序列注释。最终本研究成功鉴定出12612个树鼩基因的完整CDSs,并构建了准确度更高的树鼩基因组数据库(TupaiaBase:http://tupaiabase.org)。
