Xylem development underpins secondary growth and biomass production in woody plants. While scRNA-seq resolves early differentiation trajectories, its dependence on protoplast isolation excludes late-stage cells with thickened secondary cell walls (SCWs), leaving SCW deposition and programmed cell death (PCD) poorly characterized. By integrating snRNA-seq of Populus stem differentiating xylem (SDX) with existing scRNA-seq data, we reconstructed the full developmental continuum.
