To understand changes in gene expression profile of myeloid derived suppressor cells in tumor microenvironment and bone marrow with the shift of immune paradigm in response to progressive tumor developments. CD11b+ Gr1+ Myeloid derived suppressor cells were isolated using FACS from ascites and bone marrow from S-180 tumor bearing animals of 4 stages of tumor as 3day post tumor innoculation, 7day post tumor innoculation, 14day post tumor innoclation, 21day pos tumor innoculation which is early to late phase tumor transition and the normal counter part also isolated to compare the gene expression profile difference between control and tumor microenvironment derived MDSC. Whole cell mRNA were isolated using ambion RNA isolateion mini kit. Each group of sample represents two biological replicates (Control1, Control2), each replicate contains pulled RNA from 12 animals. 5 groups of samples as control, 3day, 7day, 14day, 21day containg one replicate in both ascites and bone marrow in total 20 samples were analysed.
