Prevotella intermedia and Prevotella nigrescens isolates genomic sequencing. Prevotella

Metagenomic sequencing helps to reveal the relationship between Prevotella and periodontal disease, but the role of subspecies diversities and genomic variations of this Gram-negative bacteria in periodontal diseases still need to be investigated. Here we performed a population-genomic analysis on the variation of Prevotella intermedia and Prevotella nigrescens isolates and its association with periodontal disease. Totally 48 Prevotella isolates were collected from different sites of 35 subjects: 14 P. intermedia isolates, derived from sites with periodontal disease (Pi-disease); 6 P. intermedia isolates from healthy sites (Pi-health); 17 P. nigrescens isolates from diseased sites (Pn-disease); and 11 P. nigrescens isolates from healthy sites (Pn-health). Genomic DNA of each isolate was extracted and pooled into 4 samples according to the species and isolated sites. sequencing libraries were built and sequenced on an Illumina HiSeq 2500 system.
