To define the composition of “first responder” lymphocytes that may engage in crosstalk with injured epithelium we profiled the lymphoytes from unwounded health skin and skin from different time points after wounding with CITE-seq and single-cell RNA-sequencing. Lymphocytes (CD45.2+ CD90.2+ TCR Vγ3 - )from unwounded skin, 3, and 5 days post wounding were sorted based on surface marker staining. Cells were prelabeled with surface epitope marking oligo-tagged antibodies and sample specific oligo-tagged Totalseq-A antibodies (Biolegend, see table 1). Hashed samples were pooled for scRNA-seq with 10X Chromium 3' v3 platform. RUN1 and RUN2 represent technical replicates.
