This database contains several images in Imaris (.ims) format obtained using lightsheet microscopy from tissue-cleared human bone biopsies. Young samples were obtained from subchondral bone from patients at 12–14 years of age undergoing epiphysiodesis. Aged (55–84 years old) human bone samples were obtained from subchondral bone from patients undergoing femoral head replacement surgery following hip replacement. Bone biopsies were stained with HCR for mRNA and immunostaining for labeling proteins. After that the samples were tissue cleared by the iDISCO pipeline. Images were taken using a LaVision ultramicroscope. "Young human bone scan"These images were taken with young human bone samples stained with YOPRO-1 for nuclei, CD31 for vasculature, CXCL12 for stromal cells and perilipin for adipocytes. Autofluorescence shows collagen-rich bone matrix. "Aged human Bone Endoglin"These images were taken with aged human bone samples stained with Endoglin for sinusoids. "Young human bone vascular rich"These images were taken with young human bone samples stained with YOPRO-1 for nuclei, CD31 for vasculature, CXCL12 for stromal cells and perilipin for adipocytes. Autofluorescence shows collagen-rich bone matrix. The region reveals a vascular niche enriched in stromal cells. "Young human bone adipose tissue"These images were taken with young human bone samples stained with YOPRO-1 for nuclei, CD31 for vasculature, CXCL12 for stromal cells and perilipin for adipocytes. Autofluorescence shows collagen-rich bone matrix. The region reveals an adipose niche enriched in stromal cells and adipocytes. "Aged human bone CD31"These images were taken with aged human bone marrow samples stained with CD31 for capillaries (shown in yellow), including filament rendering. "Young human bone sinusoidal niche"These images were taken with young human bone samples stained with CXCL12 for stromal cells, Endoglin (ENG) for sinusoids, and CD271 for mesenchymal cells.
