GEMiNI-seq for CRISPR Cas9

The GEMiNI-seq assay investigates the impact of chromatin structure on gene editor efficacy. GEMiNI-seq utilizes a library of nucleosome sequences to examine all gene-editing target locations throughout the nucleosome. Quantifying the digestion of each target generates a landscape for gene editor accessibility into the nucleosome, allowing for the most efficient targets to be selected. GEMiNI-seq tested the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) Cas9 gene editor and revealed that on-target locations within a nucleosome are less digested by Cas9 than exposed mismatched targets. The results show targeting nucleosome-bound locations will increase the probability of Cas9 editing undesirable off-targeting locations.
