Bulk MARS-seq analysis of gene expression in exocrine cells infected with Ad-PDX1, Ad-DN-REST or both. DN-REST activated endocrine genes, many of which exhibited a synergistic effect in coexpression with PDX1. PDX1 alone activated exocrine genes. Overall design: Primary exocrine cells were isolated from C57BL/6 mice and infected with Ad-mCherry control virus Ad-PDX1, Ad-DN-REST or Ad-PDX1+ Ad-DN-REST. Total RNA was obtained from 4 biological replicates 5 days post infection. This RNA was used for library construction using Bulk MARS-seq protocol and sequencing.
