We would like to investigate the level of parasitaemia in infected sandflies - initially using artificial mixtures of sandfly and parasite DNA, then moving on to sandflies artificially infected in the laboratory, but then in field-collected sandflies confirmed to be infected with Leishmania by PCR. If parasitaemia is high enough, we will validate SureSelect as an approach to extract Leishmania genomic DNA from sandflies. Secondary aims are to investigate to what extent we get sufficient sandfly genomic DNA remaining in SureSelect enriched samples to genotype the flies, and whether we can also, simultaneously identify sources of blood meals and plant food exploited by the flies from the sequencing data.
