The single-nucleus RNA sequencingq of AAV-Aβ42 injected hippocampus of macaca fascicularis

To explore the difference in Aβ42 influence between young and aged monkeys, we performed unilateral injections of AAV-Control and AAV-Aβ42 virus into the left and right hippocampi, respectively, of young (7 years) and aged (26–28 years) monkeys (4 groups, n = 3 per group). To obtain a more comprehensive infection, each hippocampus was injected at three points (anterior, middle, and caudal; 15 µL per point, 1 × 10^13 vg/mL). After three months post-injection, the left and right hippocampus tissues of the euthanized monkeys were dissected and collected separately. To collect the whole hippocampal region as completely as possible, each single hippocampus was flattened to capture the entire superficial layer and placed into a tube. The collected tissues were kept at 4°C and sent to Dynamic Biosystems Inc. (Suzhou, China) to isolate the cellular nuclei for performing the single-nucleus sequencing with the Well-Paired-Seq platform.
