An innovative coating combining polycationic poly-ε-lysine (PEL) and hyaluronic acid (HA) previously demonstrated promising in vitro results. However, subsequent in vivo results on antibacterial efficacy were inconsistent. Therefore, this study aimed to enhance the antibacterial efficacy of the coating by increasing PEL concentration twofold (from PEL-10 to PEL-20), elevating HA levels fivefold (from HA-1 to HA-5), and/or adding polyarginine (PAR30-4). These four coating formulations underwent in vitro safety and antibacterial efficacy assessments based on Safe-by-Design (SbD) principles. In vitro testing included bacterial adhesion, osteoblast behavior, and immune response evaluations.
