Using a TraDIS library produced on Klebsiella pneumoniae strain, ATCC43816, and Klebsiella pneumoniae bacteriophages RAD1, RAD2, RAD3, we aim to determine the specific receptors used to infect their bacterial host. These phage recognise capsular polysaccharides and we wish to investigate the regulatory factors and other genes responsible for Capsule biosynthesis in this Klebsiella strain. The phage will be used to eliminate mutants in the TraDIS library that have altered outer membrane profiles i.e. alterations in capsule or other outer membrane associated molecules. This methology will hopefully uncover secondry receptors that we suspect are also essential for productive phage replication and infection as we show that non-capsulated varients of ATCC43816 are still capable of being infected by RAD1,2 and 3, albiet at a lower infectivity level.
