The ribosomal RNA genes are encoded in large, tandemly repeated arrays (rDNA). Strains within a species can vary in their rDNA copy number. We used Nextera-based whole genome sequencing to genotype a set of S. cerevisiae wild isolates with a wide range of rDNA copy numbers and compared these WGS-derived rDNA copy number estimates to pulsed-field gel electrophoresis-based copy number estimates.
