Microbial DNA Enrichment from Long-term stored Sugarcane Leaves for improved Shotgun Sequencing

We present a simple approach that combines the use of a chaotrophic agent and PEG/NaCl to obtain high amounts of pure DNA from long term stored sugarcane leaf tissue. This DNA can be enriched for high-molecular weight, genomic plant DNA, or low-molecular weight microbial DNA for subsequent shot gun sequencing. Comparison of DNA derived by PEG/NaCl enrichment methods, alone or in combination with commercial, epigenetic based microbial enrichment, allows to fully cover the sequence of phytoplasma and bacterial DNA from sugarcane leaves
