We used CRISPR/Cas9 to knock in three human metabolic disease risk genes (PNPLA3I148M-GIPRdn-hIAPP) into the H11 locus of the Bama pig genome. Combined with high-fat and high-sucrose diet (HFHSD) induction, we aimed to construct a preclinical large animal model for metabolic-associated fatty liver disease (MAFLD). The triple-transgenic pigs (TG) showed hepatic steatosis, inflammatory infiltration, and fibrosis. This RNAseq study compared the 4 TG with 4 wild-type controls (WT) that were also induced by HFHSD but had no liver pathological changes, providing gene expression data for studying the molecular mechanisms of liver fibrosis in the triple-transgenic MAFLD pig model.
