Rusts are the major threat for sustainable wheat production worldwide. Considering the projected wheat demand and continueous depletion in resources, full-proof effective strategies are required. Molecular breeding approaches appeared to be successful strategy against rusts but resistance imparted is break down after any change in genetic make-up of pathogen and is frequently reported in wheat rusts. Therefore, for sustainable resistance transcriptomes based study of various factors are required in resistant and sucptible wheat plants. These factors mostly involved in: cell wall fortification, oxidative burst and production of reactive oxygen species, proteinase inhibitors, hydrolytic enzymes that degrade fungal cell wall components and pathogenesis related proteins. With this objective we mainly focused on wheat-leaf rust interaction and generated four transcriptomic SAGE libraries: susceptible-mock (S-M), susceptible-pathogen inoculated (S-PI), resistant-mock (R-M) and resistant-pathogen inoculated (R-PI) and sequenced on ABI SOLiD system 3.0.
