Practical comparison of PacBio Iso-Seq library construction approaches. undefined

With the same mouse brain RNA sample, three PacBio Iso-Seq libraries were constructed with three different protocols: 1) Standard Clontech approach (PolyA enrichment only); 2) Lexogen kit V1 (PolyA enrichment + 5'cap enrichment); 3) Lexogen kit V2 (PolyA enrichment + 5'cap enrichment). Each library was sequenced with one PacBio SMRT cell. The mouse was bred under well-controlled lab condition, with temparature of 22 celcus degree, and humidity ranging from 55-60%.
