Calcified lung nodule from 17th century mummified human remains. human lung metagenome

We performed a metagenomic pathogen screening on the whole DNA extract from a 17th century calcified lung nodule from the remains of Bishop Peder Winstrup of Lund. Following the identification of Mycobacterium tuberculosis, we created a UDG-treated library of the extract to remove damaged sites characteristic of ancient DNA, and performed an enrichment of the library for Mycobacterium tuberculosis complex (MTBC) DNA. From this enrichment, we were able to reconstruct a >100-fold coverage genome, which we used as a calibrating genome for molecular dating of the MTBC.
