Agrobacterium tumefaciens based plant transformation results in diverse genomic rearrangements. Here we utilize optical long-molecule technologies to describe the size and type of structural genomic rearrangements from four Arabidopsis thaliana Col-0 T-DNA insertion lines, SALK_059379, SALK_075892, SAIL_SEQ_232 and WiscDsLox449D11. Optical mapping of these lines identified T-DNA insertions between 27,546 bp to 236,231 bp in length, as well as a variety of intra/interchromosomal rearrangements. We further investigate the sequence content of the multi-insert lines SALK_059379 and SAIL_SEQ_232 by de novo assembly of their engineered genomes from ultra-long Oxford Nanopore MinION reads.
