Cells were isolated from adult or fetal samples, using either A2B5 or CD140 as selection marker. Human mRNA was isolated from glial progenitor cells sorted on either A2B5 (adult WM from epileptic resection) or CD140a (fetal cortex, 19-20.1 week), with four biological replicates per group. cDNA was generated with the SMARTer Ultra Low RNA kit (Clontech), and libraries were created for sequencing with the NexteraXT DNA kit (Illumina). Sequencing was performed on an Illumina HiSeq 2500v4, with all samples in one batch. An average of about 30 million 100bp reads per sample were obtained after trimming and filtering reads with Trimmomatic. Reads were aligned to the human genome with STAR, read counts for genes were generated by Feature Counts, and differential expression analysis was performed with DESeq2.
