Homo sapiens and Mus musculus Targeted Locus (Loci). synthetic construct

The goal of this study was to develop a high-throughput method of fuctionally assessing mammalian enhancer activity in embryonic stem cells and cells differentiated into more terminal cell types. ES cell libraries containing short fragments of random mouse or human BAC sequence integrated at a specific genomic locus along with a fluorescent reporter were created. Cell libraries were sorted by fluorescent activity, and the inserted DNA sequences sequenced in both highly fluorescent cell populations and unsorted library controls.
