Narcissus tazetta var. chinensis belongs to the Amaryllidaceous family and is a perennial bulbous plant. As one of the ten most famous traditional flowers in China, it is deeply loved by consumers, with its beautiful flower and rich fragrance serving high ornamental and economic value. Flower color and scent are important ornamental traits in flowers and are also main characteristics adopted for plants to attract pollinators to ensure the reproductive success of the plant. Understanding the metabolic regulatory mechanisms of growth and development, floral pigmentation and floral scent in Narcissus tazetta is an important foundation for the realisation of molecular breeding in Narcissus tazetta, and its research results have broad application prospects. In our previous studies, the transcriptomic gene expression profiles of tepals and corona at different developmental stages were used for weighted gene co-expression network analysis (WGCNA) to reveal the molecular network of metabolic associations between flower color and scent during development, and to screen some potential transcription factors (e.g. NtMYB12) involved in the regulation of flower color and scent formation in Narcissus tazetta. In this study, the NtMYB12-overexpressed and NtMYB12-RNAi transgenic narcissus callus were constructed based on the genetic transformation system of narcissus callus established in the laboratory, and their transcriptome sequencing was carried out to further study the biological function of NtMYB12 and provide a molecular basis for understanding of the formation and regulation of narcissus floral color and scent.
