Using RiboMethSeq, we systematically reinvestigate X. laevis ribosomal RNA 2’-O methylation, confirming all previously 89 mapped sites and identifying 14 novel putative positions on 18S and 28S rRNA. 23 positions are validated orthogonally by low dNTP primer extension, including 10 of the new positions.
