Radiotherapy-induced bone loss represents an increasingly important clinical challenge for which effective countermeasures are still lacking, especially considering the risk of tumor recurrence. Herein, we investigated the low-frequency electromagnetic field (PEMF) can noninvasively ameliorate radiation-induced bone damage by specifically modulating osteoblasts. We performed RNA-seq on radiated primary osteoblasts grown in osteogenic medium (MEM containing 10% FBS and osteogenic induction supplements) with or without PEMF stimulation. The total RNA of primary osteoblasts was isolated and purified using TRIzol reagent, and then reverse-transcribed to the cDNA by SuperScript Reverse Transcriptase. After amplified with PCR, the 150 bp paired-end sequencing (PE150) was performed on an Illumina Novaseq 6000 (LC-Bio Technology, Hangzhou, China) following the manufacture's protocol.
