Whole genome sequencing was performed to identify mutations created by APOBEC3B in the context of VSV resistance in a B16 murine melanoma in vitro infection model. To do this, B16tk cells stably expressing human APOBEC3B enzyme were subjected to three rounds of serial infection with vesicular stomatitis virus (VSV) expressing GFP. Cells that escaped VSV-GFP infection were harvested for genomic DNA and underwent whole genome sequencing, with B16tk parental cell genomic DNA as a control. The resulting sequences can help determine the mutational targets of APOBEC3B, which has relevance in anti-viral immunity and cancer biology.
