Alternative Splicing Events in Aedes aegypti Midgut Under Variable Temperatures: Dataset and rMATS Analysis Scripts

This dataset contains the results of an alternative splicing analysis performed on the midgut tissue of Aedes aegypti mosquitoes exposed to different temperature conditions. Alternative splicing plays a critical role in regulating gene expression and can be influenced by environmental factors such as temperature. Understanding splicing dynamics in vector mosquitoes under thermal stress is important for elucidating mechanisms of adaptation, physiology, and vector competence. Midgut tissues were dissected from adult Aedes aegypti females reared under different temperatures (20, 28 and 36°C). Total RNA was extracted, and RNA-Seq libraries were prepared and sequenced using the Illumina platform. Over two million reads per library were successfully mapped to the Ae. aegypti genome (NCBI ID: GCA_002204515.1). Alternative splicing events were identified and quantified using the rMATS (replicate Multivariate Analysis of Transcript Splicing) software. The dataset includes: Differential splicing analysis results across temperature conditions. Five major types of alternative splicing events: Skipped Exon (SE), Retained Intron (RI), Alternative 5’ Splice Site (A5SS), Alternative 3’ Splice Site (A3SS), and Mutually Exclusive Exons (MXE). Event-level statistics including inclusion level differences, p-values, false discovery rates (FDR), and junction read counts. The repository also contains the script used to execute rMATS for alternative splicing analysis.
