We used massively parallel reporter assay (MPRA) technique to identify the regulatory variants in 22 GWAS associations for the blood malignancy multiple myeloma (MM). For 1,032 variants, we designed 12 oligonucleotide sequences (2 for reference and alternative x 2 strands x 3 sliding 120-bp windows), and cloned into a vector (Tewhey et al., 2016) with ∼500 random 2- nucleotide barcodes/oligo. The library was transduced into L363 MM cells in 3 replicates. Reporter mRNA was sequenced.
