The invention discloses application of GhPRXR1 protein and coding gene thereof in regulating and controlling oil content of cottonseed. The invention clones cotton GhPRXR1 gene from upland cotton material Msco-12， successfully constructing yeast over-expression vector and transforming saccharomyces cerevisiae. Compared with the trans-empty vector yeast control， the oil content of the 3 positive transgenic yeasts is respectively improved by 20.01%， 34.79% and 37.25%. The invention also successfully constructs a virus-induced GhPRXR1 gene silencing vector and transforms upland cotton Msco-12. The average oil content of 6 GhPRXR1 gene silencing cotton positive individual mature cottonseed is reduced by 18.11% compared with that of 3 control individual mature cottonseeds， indicating that the GhPRXR1 gene can regulate and control oil content of cottonseed， having great value in improving the cottonseed yield and cotton breeding.
