Transcriptome sequencing of dorsal and subventral glands from mixed life stage pine wilt nematodes, Bursaphelenchus xylophilus

Transcriptome sequencing of total RNA isolated from approximately 200 mixed dorsal and subventral glands microaspirated from mixed life stage pine wilt nematodes, Bursaphelenchus xylophilus, cultured on Botrytis cinerea in flask for 7 days at 25C. Total RNA was isolated using the Arcturus PicoPure RNA isolation kit (Thermofisher Scientific) yielding approx 4 ng.This was amplified using the SMARTer Stranded Total RNA-Seq kit - Pico Input Mammalian (Clontech, USA), ribosomal cDNA was depleted, and after a final PCR amplificationlibraries were sequenced. Quality of RNA and cDNA assessed using a Bioanalyzer. Two paired-end libraries (technical replicates BX-1 and BX-2 from the same pooled biological sample) were sequenced using the NextSeq Illumina service from Admera Health (USA).
