RNA-seq of BJ fibroblasts transfected with control or FH siRNA

Bulk RNA sequencing was performed on BJ human fibroblasts transfected with control siRNA or FH-targeting siRNA to investigate transcriptomic changes associated with FH silencing. Four biological replicates per condition were analyzed. Overall design: The RNAseq sequencing was performed by the GenomIC Cochin Institute INSERM U1016. BJ cells were transfected (siC vs. siFH) and cultured for 5 days before RNA extraction (4 samples per condition). Following RNA quality evaluation, libraries were prepared using the Stranded mRNA Prep protocol from Illumina, starting from 250 ng of total RNA. Paired end (2 × 51 bp) sequencing was performed on an Illumina NovaSeq platform.
