Epigenetic profiles of HSCs transduced with control or Runx3-expressing lentivirus

To determine if ectopic expression of Runx3 can correct deficient lymphoid output in aged HSCs, we examine the chromatin profile of aged HSC transduced with control or Runx3-expressing lentivirus. Control or Runx3-expressing lentivirus was used to infect cKit-enriched bone marrow cells from aged (24 months) mice. Transduced cells were transplanted into lethally irradiated mice. After 16 weeks of transplantation, transduced donor-derived HSCs (LSK Flt3-CD150+CD48-CD45.2+hCD2+) were FACS sorted for ATAC-seq profiling.
