Raw data files used in volume correlative light X-ray and electron microscopy (CLXEM) workflow performed for the study "Autophagy modulates endothelial junctions to restrain neutrophil diapedesis during inflammation" by Reglero-Real et al. (Immunity 2021). Mice expressing Cre-recombinase under the endothelial cell specific Cdh5 promoter, the Rosa-26-Tomato Cre reporter and floxed alleles for the autophagy related protein 5 (Atg5) were used. This mouse line exhibits wild type (tdTomato-) and Atg5-/- (tdTomato+) endothelial cells in a mosaic manner. Cremaster muscle microcirculation was first labeled in vivo with anti-mouse PECAM-1 antibody conjugated with Alexa Fluor 488. Next, the cremaster muscle was dissected, fixed with formaldehyde and imaged using Zeiss LSM800 prior to the sample preparation for electron microscopy.
