This study aimed to investigate the relationship between alternative splicing and intrinsic tumor radiosensitivity in non-small cell lung cancer (NSCLC) cells by integrating second- and third-generation sequencing technologies. We performed transcriptome analyses on the H1299 cell line and its radioresistant derivative, H1299R, before and after irradiation (5Gy, 12 hours). Our analysis identified differentially expressed genes and transcripts and revealed dynamic changes in splicing events induced by irradiation over time. Notably, irradiation significantly reduced the number of transcripts generated by genes, while the total number of alternative splicing events showed a relatively smaller decrease.Furthermore, we identified key splicing factors related to radiotherapy prognosis, including DHX15, PSKH1, RNPC3, and THOC1. These findings provide critical insights for enhancing radiotherapy strategies and identifying novel therapeutic targets. The data from this study contribute to a deeper understanding of the role of alternative splicing in tumor radiation response, underscoring their significant clinical relevance.
