AAV-PHP.S-cTnT-hTRPV1 cardiac transduction and long-term systematic safety dataset in mice.

This dataset contains systemic safety and cardiac transduction efficiency data following a single intravenous (jugular vein) injection of AAV-PHP.S vectors carrying the human TRPV1 channel under the cardiac troponin T promoter (pAAV_cTnT_hTRPV1_3×FLAG) at a dose of 6×10^11 viral genomes per mouse. Injections were performed in adult wild-type mice (8–12 weeks old), with a cohort of uninjected wild-type mice serving as controls. At 4 weeks post-injection, the proportion of transduced cardiomyocytes in the heart was quantified by flow cytometry. At 12 months post-injection, long-term safety was assessed by hematology and clinical biochemistry of blood. Also we performed histopathological examination of heart, liver, kidneys, and spleen to identify any pathological changes induced by transduction. The dataset provides paired comparisons between TRPV1-transduced and uninjected wild-type mice, supporting the evaluation of organ specificity and the absence of adverse off-target effects after systemic AAV-mediated cardiac gene delivery. All data are associated with the study “Thermogenetics for cardiac pacing”.
