This synthetic rumen microbiota DNA reference standard contains 13 bacteria and three archaea 16S rRNA sequences and nine protozoa 18S rRNA sequences. Amplicon sequences were generated using the Caporaso et al. (2011) primers targeting the V4 region of the 16S rRNA gene. Protozoa sequences were included to test primer specificity. The aim of the study was to investigate differences in the performance of four microbial DNA reference databases (RDP, SILVA, GTDB and RefSeq +RDP) for rumen microbiota investigations.
