Transcriptome analysis of Field Resistances to spirodiclofen in Panonychus citri (McGregor)

Spirodiclofen is one of the most widely used acaricides in China.The citrus red mite, Panonychus citri (McGregor) (Acari: Tetranychidae), is an important citrus pest around the world that has developed high resistance to spirodiclofen.In this study, we identified a field spirodiclofen-resistant strain (DL-SC) and conducted transcriptome sequencing of resistant and susceptible strain to identify candidate genes that might contribute to spirodiclofen resistance in P.citri. Compared with a susceptible strain, the field-collected DL-SC population showed 712 fold resistance to spirodiclofen by egg bioassay. Amplification and sequencing of the ACCase and CPR genes between resistant and susceptible strains did not find common non-synonymous mutations. Transcriptome expression data and quantitative real-time PCR (qRT-PCR) revealed the significantly overexpression specifically of two cytochrome P450 genes (CYP385C10 and CYP406A1). Our results suggest that the overexpression of P450 genes may be involved in the spirodiclofen-resistance of field population of P.citri in China.
