GERMIN3 has previously been identified as a downstream target of the potato tuberigen activation complex (Zhang et al. 2020; Plant Journal 103:2263-2278). In this experiment, day-length dependent Solanum tuberosum subsp. andigena wild-type and a GERMIN3 overexpression line were grown for six weeks in a glasshouse under tuber noninductive long day conditions (16h daylength 20/15oC day/night) and the apical and hook region from elongating stolons of four replicate plants of each genotype were harvested and snap frozen. Plants were transferred to inductive conditions in a growth room (10h daylength, 20/15oC day/night) for two weeks and the swelling regions of early swelling stolons (< 3mm) from four replicate plants of each genotype was harvested and snap frozen. RNA was extracted and sequenced using an Illumina NextSeq2000.
